Glucosamine is a natural compound found in cartilage, the tissue that cushions the shock and pressure exerted on joints. In supplement form, glucosamine is obtained from shellfish and crustaceans. There are several forms of glucosamine, including glucosamine sulphate, glucosamine hydrochloride and N-acetyl glucosamine. Oral glucosamine sulphate is used to treat a painful condition caused by inflammation called osteoarthritis.

Chondroitin is a natural substance found naturally in the human body. It is a major component of cartilage. The form of chondroitin is called chondroitin sulphate and is a combination of chondroitin and mineral salts that helps improve its absorption. Chondroitin helps build connective tissue throughout the body, including that which forms joints and the gastrointestinal tract. Because it works by retaining water, the benefits of chondroitin include improved lubrication and flexibility of stiff or worn tissues and joints.

Osteoarthritis is the most common type of arthritis and degenerative joint disease characterized by cartilage degradation and joint pain. Chondroitin sulphate is commonly used to treat the pain associated with osteoarthritis, especially forms that affect highly susceptible parts of the body such as the knees and hands. In general, studies have shown that the use of chondroitin leads to relief of joint pain over several months, although some people experience even more benefits.

According to the Arthritis Foundation, on average, study participants typically experience about a 10% improvement in pain symptoms when using chondroitin compared to placebo. The best results are usually obtained after using a product containing chondroitin for three months or more. 

Recently, the University of Utah School of Medicine conducted the largest clinical trial investigating the effects of chondroitin and glucosamine, called the ,,Glucosamine/Chondroitin Arthritis Intervention Trial (GAIT)". According to reports published by the National Center for Complementary and Integrative Health, GAIT is the first large-scale, multicenter clinical trial in the U.S. to test the effects of dietary supplements containing glucosamine and chondroitin for the treatment of knee osteoarthritis. 

The GAIT study compared the effects of glucosamine and chondroitin sulphate (used separately and also in combination) with the effects of a placebo and also a prescription drug. 16 rheumatology research centres in the US and over 1,500 patients participated in the study, which lasted six months. Patients received one of five treatments over six months, including the use of glucosamine and chondroitin, celecoxib (a prescription drug used to manage osteoarthritis pain) or a placebo. A positive response to any treatment was defined as a 20% or greater reduction in pain after six months compared to the start of the study.

Results from the GAIT study showed that for participants with moderate to severe pain, glucosamine combined with chondroitin sulfate provided significant pain relief compared to placebo - about 79% had a 20% or greater reduction in pain compared to about 54% for the placebo group. Chondroitin and glucosamine actually worked for more people than prescribed - 70% of participants in the celecoxib group experienced pain relief compared to placebo.
